[H-3] (+)-MK-801

MK-801 (dizocilpine) is a potent synthetic anticonvulsant and dissociative compound that acts as a pore-blocking blocker of the N-methyl-D-aspartate (NMDA) receptor. It binds to a specific binding site located deep inside the ion channel pore of the open receptor, preventing the influx of calcium (Ca2+) and sodium (Na+) ions necessary for neuronal depolarization.

MK-801 is non-competitive (or uncompetitive/use-dependent) because it does not compete with endogenous glutamate or glycine at their extracellular agonist binding sites. Instead, it requires the receptor channel to physically open first - triggered by glutamate/glycine binding - before MK-801 can enter the open pore and trap itself inside to block ion flow.

The active (+)-enantiomer of MK-801 exhibits sub-nanomolar to low-nanomolar affinity for the NMDA receptor pore, with reported binding affinity (Ki) values typically ranging from 30.5 to 37.2 nM in central nervous system membrane preparations.

In preclinical neuroscience, MK-801 is primarily used to induce schizophrenia-like behavioral and cognitive deficits in rodents to test potential antipsychotic drugs. It is also widely employed as an experimental tool to study glutamate toxicity, synaptic plasticity (such as long-term potentiation, or LTP), memory consolidation, and neuroprotection against ischemic neuronal damage.

While all three share the same binding site within the NMDA receptor pore, MK-801 binds with significantly higher affinity and has a much slower dissociation rate, causing an extremely prolonged and almost irreversible channel block. Consequently, MK-801 exhibits far greater potency and long-lasting behavioral effects, making it too toxic and neurodisruptive for clinical human use compared to ketamine.

In rodents, low to moderate doses of MK-801 produce intense hyperlocomotion, ataxia, stereotyped behavior (head-weaving, turning), and severe cognitive impairments in memory tasks. These behaviors closely model both the positive symptoms (psychomotor agitation) and negative/cognitive deficits observed in human schizophrenia.

MK-801 is a dangerous neurotoxin that can cause Olney's lesions (neuronal vacuolization) in rodent brains, severe central nervous system disruption, and potent psychoactive effects. Handling requires strict laboratory containment, chemical safety goggles, gloves, and a chemical fume hood to prevent accidental inhalation, ingestion, or dermal exposure.
