[H-3] TBOB

t-Butylbicycloorthobenzoate (TBOB) is a synthetic bicyclic cage compound and potent neurotoxin used in neuroscience research. It acts as a convulsant by binding to the picrotoxin site inside the GABAA receptor chloride channel, directly blocking the influx of inhibitory chloride ions.

TBOB binds inside the central pore of the GABAA receptor ion channel, directly blocking the passage of chloride ions. Access to this picrotoxin-sensitive binding site requires activation by GABA, which opens the channel pore and allows TBOB to insert and trap the receptor in a non-conducting state. Its hydrophobic $t$-butyl group and aromatic phenyl ring anchor the molecule in place, producing a slow dissociation rate that causes prolonged channel block.

TBOB acts as an uncompetitive or non-competitive channel blocker that directly binds inside the open GABAA receptor pore to physically obstruct ion passage. As a result, TBOB significantly reduces or completely abolishes GABA-induced inward chloride currents without altering GABA's ability to bind to its primary agonist site. This blockage stabilizes the receptor in a non-conducting state, preventing hyperpolarization and leading to sustained neuronal hyperexcitability.

H3-TBOB is primarily used in radioligand binding assays and quantitative autoradiography to map and characterize the picrotoxin/convulsant binding site within the GABAA receptor chloride channel across brain tissue. Researchers also employ it to investigate allosteric channel modulation by drugs, neurosteroids, anesthetics, and insecticides, as well as to evaluate changes in receptor density during conditions like epilepsy or chronic alcohol exposure.

TBOB and TBPS differ primarily in their chemical cores—a phenyl-containing orthoester versus a thiophosphoryl group—which alters their channel dissociation rates and lipophilicity. Additionally, TBOB is typically tritium-labeled for a longer shelf life and higher assay stability, whereas TBPS is commonly labeled with sulfur-35 as the historical reference standard.

TBOB and related convulsant ligands present severe safety risks because they are potent GABAA receptor antagonists capable of triggering violent seizures, respiratory failure, or death even at microgram doses. Exposure occurs rapidly via inhalation of dust, accidental needle sticks, or dermal absorption through organic solvents like DMSO. Safe handling requires strict engineering controls like certified fume hoods, double-gloving, and secure hazardous waste disposal.
