[H-3] DPCPX

DPCPX (1,3-dipropyl-8-cyclopentylxanthine) is a synthetic, cell-permeable xanthine derivative that acts as a potent xanthine-based antagonist at adenosine receptors. It functions primarily as a highly selective competitive antagonist at the A1 adenosine receptor subtype across central and peripheral tissues.

DPCPX demonstrates exceptional selectivity for the A1 receptor, exhibiting over 500-fold to 700-fold higher affinity for A1 over A2A receptors. It shows even greater selectivity over A2B and A3 receptor subtypes, making it one of the most reliable pharmacological tools for selectively blocking A1-mediated signaling.

At human recombinant adenosine receptors, DPCPX binds with sub-nanomolar affinity to A1 (Ki = 0.45-1.3 nM). In contrast, its affinity is significantly lower at A2A (Ki = 130-340 nM), A2B (Ki = 30-100 nM), and A3 (Ki > 1000-10000 nM).

DPCPX is widely used in neurophysiology to study A1-mediated presynaptic inhibition, neurotransmitter release, and synaptic plasticity like long-term potentiation. Radiolabeled variants (3H-DPCPX) are also routinely employed in receptor binding assays and autoradiography to map A1 receptor density in brain tissue.

Unlike caffeine and theophylline, which are weak, non-selective adenosine receptor antagonists that block A1 and A2A receptors with roughly equal, micromolar affinity, DPCPX displays sub-nanomolar affinity and remarkable subtype selectivity for the A1 receptor. This extreme selectivity allows researchers to isolate A1 actions without triggering broad off-target adenosine blockade.

While DPCPX itself served as a prototype for selective A1 blockade, its poor aqueous solubility and potential for off-target effects prevented direct clinical approval. However, structural modifications of the DPCPX scaffold guided the development of related xanthine and non-xanthine A1 antagonists investigated as potential diuretic, renal-protective, and cognition-enhancing drug candidates.
